Alu/PCR Lab
Objectives - Successfully isolate DNA from cheek cells, prepare a PCR reaction for amplification of an Alu insert.
Materials -
.9% saline solution
micropipette tips
waste container
microcentrifuge
microcentrifuge tubes
PCR tubes
agarose
1x TAE
gel chambers + molds
load dye
chelex
racks
primer mix
master mix
H20
+ control DNA
Procedure - We took DNA from inside our mouths with salt water around and put that into a tube. We got rid of the top stuff, all but the cell pellet at the bottom. Then we added saline and 5% chelex. After we put our tube into a heat block for 10 minutes. After spinning, we took out the portion of liquid at the top, not the chelex beads at the bottom into a new tube. We took master mix and put it into the small PCR tube, also adding primer mix and a smaller amount of DNA. Lastly once it was spun, we pipetted our mix into the agarose gel. After the electrophoresis was completed it will be photographed.
